Peptide Nasal Sprays

Nasal administration refers to a method of delivering drugs through the nasal passages. This approach can serve as either a topical or systemic administration, as the drugs delivered locally may exert either local or systemic effects. Peptide nasal sprays typically consist of locally acting medications, such as decongestants used for treating colds and allergies, which generally have minimal systemic effects. Conversely, examples of systemically active drugs available in nasal spray form include medications for migraines, nicotine replacement therapies, and hormone treatments.

Limitations with nasal systemic drug delivery

Nasal administration is mainly appropriate for potent drugs, as only a limited volume can be introduced into the nasal cavity. Medications intended for continuous and frequent use may be less suitable due to the potential for adverse long-term effects on the nasal epithelium. Additionally, nasal administration has been linked to considerable variability in drug absorption. Factors such as upper airway infections can increase this variability, as can the degree of sensory irritation of the nasal mucosa, variations in the volume of liquid spray that is swallowed rather than retained in the nasal cavity, and differences in the spray actuation technique. Nevertheless, the variability in absorption following nasal administration should be considered.

Advantages of nasal systemic drug delivery

The nasal cavity is lined with a thin, well-vascularized mucosa. Consequently, drug molecules can be rapidly transferred across the single layer of epithelial cells directly into the systemic blood circulation, bypassing first-pass hepatic and intestinal metabolism. The onset of effects is often observed within five minutes for smaller drug molecules. Thus, nasal administration can serve as an alternative to oral routes, where tablets or capsules may be crushed or ground into a powder and then snorted or sniffed, leading to a swift onset of effects. This method is particularly beneficial when a rapid effect is required or when the drug is significantly degraded in the gastrointestinal tract or liver, allowing for the administration of drugs that are poorly absorbed orally.

Storage Recommendation

All peptide products, whether mixed or unmixed, should be kept in the refrigerator (at temperatures between 4 and 20 degrees Celsius), instead of the freezer. Furthermore, peptides can be stored in a cool, dark, and dry place, away from direct sunlight.

A nasal cannister should be used 2 squirts per nostril per day, a total of 4 squirts. The nasal spray will last 15 days as it contains 60 sprayer activations.
